This method uses the bacterial enzyme polynucleotide phosphorylase, which is specific for ribonucleotides, but polymerizes deoxyribonucleosides albeit at a slow rate when Mn2+ replaces Mg2+ in the reaction mixture. It uses as substrate deoxyribonucleosides diphosphates and does not require a template. But it does need a primer at least 3 nucleotides long. The enzyme adds one or two nucleotides at a time to the primer (and to the growing chain).
